Total chlorine determination in water quality analysis relies heavily on the DPD colorimetric method. The reagents involved—particularly DPD indicator, buffer solutions, and oxidising agents—are chemically active and sensitive to environmental conditions.
Improper storage can degrade them rapidly, leading to biased results or complete test failure. Therefore, a systematic storage protocol is essential for maintaining reagent integrity and analytical reliability.
Temperature Control
Temperature is the primary factor governing reagent stability. Liquid DPD reagents are typically required to be stored under refrigeration at 4 ± 4 °C, in closed, light‑tight containers. Some ready‑to‑use liquid reagents specify a storage range of +6 to +10 °C. Solid reagents, such as powders and tablets, are generally stored at ambient temperature, provided the environment is cool (below 25 °C), dry, and dark.
Before use, all reagents must be brought to room temperature—usually 20 ± 3 °C—and thoroughly shaken or mixed. This equilibration ensures accurate pipetting and consistent reaction kinetics.
Light Protection and Airtight Sealing
DPD and other chromogenic agents are photosensitive. Ultraviolet and visible light accelerate decomposition, causing discolouration and loss of reactivity. Reagents should be kept in amber glass bottles or opaque packaging, and never transferred to clear containers.
Sealing is equally critical. Oxygen, moisture, and airborne contaminants can oxidise or hydrolyse the reagent. After each use, the bottle cap must be tightly replaced immediately. Never return unused reagent to the original container to avoid cross‑contamination.
Moisture Prevention for Solid Reagents
Powders and tablets are hygroscopic. Exposure to humid air causes caking, degradation, and reduced active content. They must be stored in a dry, well‑ventilated location, ideally with desiccants. Foil‑pouched unit‑dose reagents offer excellent moisture protection but should be used promptly after opening.
Shelf Life and Expiry Management
The usable lifetime varies by formulation. DPD liquid reagents generally have a shelf life of about 2 years when stored correctly. Ready‑to‑use DPD reagent packets for field testing typically expire within 6 months. Tablets can last up to 10 years due to their solid, stable form. For online analyser reagent kits (e.g., CL17 series), the stated shelf life is often 6 months. Some prepared standard solutions remain stable for only 1 day under refrigeration.
For expired reagents, manufacturers may indicate that they can still function if verified regularly with standard solutions. A simple check—measuring a standard near the upper range of the test—can confirm whether the reagent still performs accurately across the working range. However, once proven ineffective, they must be disposed of according to safety data sheets and laboratory waste guidelines.
Operational Best Practices
When using reagents, avoid contamination. Use clean, dry spatulas for solids. Do not touch the inside of caps or bottle necks. Open ampoules must be used immediately and cannot be resealed for future use. Always follow the manufacturer’s instructions regarding storage orientation, shaking, and the order of addition.
