    The determination of Biochemical Oxygen Demand (BOD) is one of the most critical parameters for assessing organic pollution in water and wastewater. However, the accuracy of BOD results is highly dependent on proper sample handling from the moment of collection. Among the most frequently asked questions in water quality analysis is: how long can a BOD sample be stored before analysis?

    The answer is not a single number, as different regulatory and standard-setting bodies offer varying guidelines. Nevertheless, a clear consensus emerges: the sooner the analysis begins, the better – and under no circumstances should a BOD sample be held for more than 48 hours.

    The 24-Hour Standard

    The most widely accepted benchmark comes from Standard Methods for the Examination of Water and Wastewater (Method 5210B), which explicitly states that the maximum holding time for BOD samples is 24 hours. Samples should be cooled to near-freezing temperature (approximately 4°C) immediately after collection to suppress microbial activity, and then analyzed within this 24-hour window. If analysis can begin within two hours of collection, refrigeration is not even necessary.

    The 48-Hour Regulatory Allowance

    In the United States, the Code of Federal Regulations (40 CFR Part 136) permits a longer holding time of 48 hours. However, this is clearly stated as a maximum – not a recommendation. The footnote accompanying this regulation emphasizes that samples should be analyzed as soon as possible after collection, and the 48-hour limit should not be interpreted as a license to delay analysis.

    The Practical 6-Hour Guideline

    For the most reliable results, many environmental agencies and laboratories recommend an even stricter timeline. The U.S. EPA and various state departments advise that BOD analysis should begin within 6 hours of sample collection whenever feasible. When samples must be transported to an off-site laboratory, every effort should be made to meet the 24-hour holding time; if logistics make this impossible, samples should be shipped the same day and analyzed as early as possible the next day.

    Special Cases: Chlorinated Samples

    An important exception exists for samples containing residual chlorine. In such cases, the sample must be dechlorinated at the time of collection, and the maximum holding time can be extended to 28 days. This is because the dechlorination process stabilizes the sample and prevents ongoing chemical interference with the BOD measurement.

    Why Holding Time Matters

    The rationale behind these strict time limits is straightforward: BOD measures the oxygen consumed by microorganisms as they degrade organic matter. This biological process begins the moment the sample is collected and continues – albeit more slowly – even under refrigeration. Delaying analysis allows this consumption to progress, resulting in BOD values that no longer reflect the true oxygen demand of the original sample.
